Flow Cytometric DNA Content Analysis in Endometrial Hyperplasia and Endometrial Adenocarcinoma



Abstract
T o evaluate the usefulness of DNA ploidy and cell cycle study in the distinction between endometrial hyperplasia and adenocarcinoma as well as in the assessment of the aggressivenss of mallignancy, flow cytometric analysis of DNA content was
done
in
paraffin-embedded tissue sections of 10 normal, 32 hyperplastic, and 20 adenocarcinomatous endometria, which were diagnosed and treated in Korea University Hospital.
@ES The obtained results were as follows:
@EN 1. Aneuplod DNA pattern was only found in 3 of 20 endometrial adenocarcinoma (15%).
2. S-phase fraction in adenocarcinoma was significantly increased than in normal and hyperplastic endometrium.
3. Proliferative index(S+G2/M phase) was significantly increased in adenomatous hyperplasia than in endometrium of normal and simple hyperplasia, and also in adenocarcinoma than in adenomatous hyperplasia.
4. S-phase fraction and proliferative index (S+G2/M phase) ware significantly increased in poorly differentiated adenocarcinoma than in well to modrately differenciated adenocarcinoma.
5. Three cases of adenocarcinoma with DNA aneuplody were in clinical stage III or IV and histologic grade 2 or 3.
From the above results, it is condulded that flow cytometirc DNA conctent analysis can be used in differentiation between endometrial hyperplasia and adenocarcinoma, and in evaluation of the prognosis of malginancy.
